My personal favorite is:
Umbridge: Potter has received very poor marks in all his tests with me.
McGonagall: I should have made my meaning plainer. He has achieved high marks in all Defense against the Dark Arts tests set by a competent teacher.
Here are a few other gems I haven't seen people mention:
Hermione (while blackmailing Rita Skeeter): I suppose the Prophet would pay you rather a lot for an insider's account of Azkaban....
McGonagall (asked if she and the other professors can secure the school against Voldemort): Yes, I think so. We teachers are rather good at magic you see.
Ron (about Aragog): Believe me, being dead will have improved him a lot.
Phinneas Nigellus Black: I disagree with Dumbledore on many counts, Minister, but you can't deny he's got style. (This was modified and given to Kingsley Shacklebolt in the movie)
Edit: My original quote from Ron was actually said by Harry (and mentioned previously), so I've replaced it with a gem he actually did say. McGonagall's answer was slightly off, so I edited that, too
